چکیده

Macrocyclic ligands 1 and 2 containing two positively charged phenanthridinium units and aminobisacetylenic bridges, exhibit significantly higher affinity toward single-stranded rather than double-stranded polynucleotides. The ligands bis-intercalate into the former and show non-intercalative interactions with the latter type of nucleic acids. Both ligands differentiate AMP from the GMP or UMP by significant fluorescence emission increase upon complexation of the first nucleotide in water (log Ks of 1:1 and 1:2 complexes 5.8 and 1.4, respectively), while only a slight emission change is observed in titrations with GMP and UMP. © 2001 Elsevier Science Ltd. Novel Pt(II) Complex and Its Pd(II) Aanalogue. Synthesis, Characterization, Cytotoxicity and DNA-interaction

The ability of small molecules to perturb the natural structure and dynamics of nucleic acids is intriguing and has potential applications in cancer therapeutics. This work reports the synthesis, characterization, cytotoxicity and DNA-binding studies of two cytotoxic and intercalative [M(bpy)(pyrr-dtc)]NO3 complexes (where M = Pt(II) and Pd(II), bpy = 2,2´-bipyridine and pyrr-dtc = p...

The role of tyrosine in the association of proteins and nucleic acids. Specific recognition of single-stranded nucleic acids by tyrosine-containing peptides.

Oligopeptides containing tyrosyl, lysyl, and alanyl residues bind to polynucleotides and nucleic acids as shown by proton magnetic resonance, fluorescence spectroscopy, and difference absorption spectroscopy. Proton magnetic resonance data indicate that stacking of tyrosyl residues with nucleic acid bases takes place only in single-stranded structures (such as poly(A) or denatured DNA). Stackin...

Nucleic acid fluorescent probes for biological sensing.

Nucleic acid fluorescent probes are playing increasingly important roles in biological sensing in recent years. In addition to the conventional functions of single-stranded DNA/RNA to hybridize with their complementary strands, affinity nucleic acids (aptamers) with specific target binding properties have also been developed, which has greatly broadened the application of nucleic acid fluoresc...

Screening for oligonucleotide binding affinity by a convenient fluorescence competition assay.

A competitive homogeneous quenched fluorescence assay system is described for the high throughput screening of DNA conjugates that bind to single-stranded DNA. Fluorescence signal is generated by competitive binding of the sample molecule to a target strand labelled with a quencher probe, which is otherwise hybridised to a complementary strand containing a fluorescent probe. Thus fluorescence g...

Purification and characterization of the coliphage N4-coded single-stranded DNA binding protein.

We have purified and characterized a single-stranded DNA binding protein (N4 SSB) induced after coliphage N4 infection. It has a monomeric molecular weight of 31,000 and contains 10 tyrosine and 1-2 tryptophan amino acid residues. Its fluorescence spectrum is dominated by the tyrosine residues, and their fluorescence is quenched when the protein binds single-stranded DNA. Fluorescence quenching...
